Help with mysql (indexes?)

I have a very simple Mysql setup, and very simple Select X from Y where Z =asd; query is taking 2 seconds (2.5 mil rows)
And the server (running on windows) has very high CPU.
Using the "explain" functionality I have discovered that index is not being used, and not sure how to set it up
